Functional characterization of mosquito genes involved in dengue virus transmission identified by RNA-seq
A 3-year postdoctoral position, funded by the Laboratory of Excellence Integrative Biology of Emerging Infectious Diseases (LabEx IBEID: http://www.pasteur.fr/labex/ibeid), is available in 2015 in the Insect-Virus Interactions group led by Louis Lambrechts at the Institut Pasteur in Paris (Lab website: http://www.thelambrechtslab.net).
Dengue is the most rapidly expanding, globally dominant arthropod-borne (arbo) viral disease. Despite the critical role of arthropod vectors in arboviral emergence, the genes controlling the level of vector competence for dengue viruses in the mosquito Aedes aegypti are still unknown. The aim of this project is to take advantage of a recently developed genome-wide transcriptomic approach based on RNA-seq at the individual mosquito midgut level to identify and validate candidate genes of Ae. aegypti vector competence for dengue viruses, and proceed with their functional characterization. The project will rely on fieldderived mosquitoes and dengue virus isolates, high-throughput sequencing, statistical analysis of gene expression data, functional assays in vivo, and concepts in population biology. Results from this project are expected to provide important insights into the molecular mechanisms underlying vector-virus compatibility and insect antiviral immunity.
Candidate requirements: Applicants must hold a PhD in a relevant field and have demonstrated expertise in functional genomics and a strong background in molecular biology, confocal microscopy and basic protein biochemistry. Additional experience in virology and/or mosquito biology is desirable but not necessary. English proficiency (oral and written), excellent communication skills and ability to work collaboratively are essential. Knowledge of the French language is not required. There is no age or nationality restriction.
